294 Abnormal collagen I structure and protein expression in desmoplastic cutaneous squamous cell carcinoma

چکیده
Desmoplasia is a highly aggressive histopathologic feature of cutaneous squamous cell carcinoma CSCC independently associated with death from CSCC. It the deposition abnormal collagen around tumor as seen on H&E. However, structure this unknown. Herein, early immunofluorescence (IF) studies to define differences between normal and desmoplastic structures are reported. Tumor samples 27 patients were graded for desmoplasia using semi-quantitative grading scale. Samples prominent versus no analyzed bulk RNA-seq analysis by Nanostring technology. Additionally, 7 skin multispectral IF. K14 identified keratinocytes, vimentin (VIM) fibroblasts. COL1A1 was significantly upregulated gene in CSCC, compared non-desmoplasia (Log2FC=2, p<0.001). COL1 IF intensity decreased stroma (p<0.0001) (p<0.0001). The staining pattern showed markedly altered I comprised thin, wispy strands, keeping H & E appearance collagen. Surprisingly, increased (keratinocyte) regions CSCCs non-desmoplastic epidermis (P<0.0001). degeneration present some cells. This COL1-positive phenotype number cells/mm2compared (413 vs 1101, p<0.0001). These findings indicate that expression quantifiably structurally different native collagens. Giant desmoplastic cutaneous squamous cell carcinoma of the gluteal region
Cutaneous squamous cell carcinoma (cSCC) is the most common type of skin tumour with the ability of metastatic spread. It represents about 20% of all malignancies diagnosed worldwide each year. Despite increased knowledge regarding the causes of skin cancer, the incidence of cSCC rises. The disease originates from epidermal keratinocytes, but it may occur on all areas of the body. It has an inv...
متن کاملTissue eosinophilia in oral and cutaneous squamous cell carcinoma and normal oral and cutaneous tissues
Introduction: Eosinophils produce proteases that stimulate invasion in cancers. Due to the more malignant potential of oral squamous cell carcinoma (OSCC) compared to cutaneous squamous cell carcinoma (CSCC), the aim of the present study was to compare tissue eosinophilia between OSCC and CSCC samples to understand whether tissue eosinophilia play a role in different biological...
متن کاملThe Usefulness of CD10 in Distinguishing between Cutaneous Basal Cell Carcinoma and Squamous Cell Carcinoma
Background & Objectives: CD10 is a cell surface enzyme with metalloendopeptidase activity, also known as Common Acute Lymphoblastic Leukaemia Antigen, which mainly serves as a marker for acute lymphoblastic leukemia (ALL). To date and to the best of our knowledge, only few comparative immunohistochemical studies have assessed CD10 expression in cutaneous epithelial neoplasms. Our goal was to d...
متن کاملCytodiagnosis of Cutaneous Basal and Squamous Cell Carcinoma
Background and Objective: Cutaneous cytology is the study of scraping or smears obtained from skin lesions. Although not commonly employed, cutaneous cytology has been a useful tool for clinicians due to simplified procedures and high sensitivity and specificity of presurgical clinical diagnosis of commo...
متن کاملValue of CD10 Expression in Differentiating Cutaneous Basal from Squamous Cell Carcinomas and Basal Cell Carcinoma from Trichoepithelioma
Background: In addition to the well-defined histological criteria for squamous cell carcinoma (SCC) and basal cell carcinoma (BCC), immunohistochemical techniques can be used in difficult cases for their differentiation. As differential diagnosis between trichoepithelioma (TE) and BCC is sometimes difficult for the clinician and the pathologist, CD10 may be a useful marker for definite diagnosi...
